技術(shù)服務(wù)描述

針對目標區域設計特異性DNA探針并經(jīng)過(guò)脫硫生物素標記，脫硫生物素探針可以和偶聯(lián)在磁珠上的鏈霉親和素親和結合。然后，細胞核提取物與磁珠-DNA探針?lè )跤?，作用蛋白質(zhì)分子可以和DNA探針特異性結合；經(jīng)過(guò)洗滌可以將非特異性結合蛋白質(zhì)去除；最后，經(jīng)洗脫液洗脫，得到目的DNA探針-蛋白質(zhì)復合物，再經(jīng)過(guò)Western Blot或質(zhì)譜（MS）鑒定蛋白質(zhì)類(lèi)型。

Biotin-14-CTP is a CTP analog with biotin attached at the N 4 -position of the pyrimidine base by a 14-atom linker. Biotin-14-CTP can be efficiently incorporated into RNA transcripts using T7, SP6, or T3 RNA polymerase and the appropriate template in the presence of ATP, GTP, UTP, and Biotin-14-CTP (1). The biotinylated RNA can be used as a probe in membrane-based or in situ hybridizations (2). The amount of material provided is sufficient for up to 20 transcription reactions (1 μg template).
